| Abstract | Banurea, Malumta. Formulation and Evaluation of the Antibacterial Activity of a Hand Sanitizer Containing Mangosteen (Garcinia mangostana L.) Leaf Extract Against Escherichia coli. Thesis, Department of Pharmacy, Faculty of Pharmacy, Deli Husad Deli Tua Health Institut. (Supervised by : apt. Sulasmi, S.Farm., M.K.M, M.Farm) Hand hygiene is one of the most important measures to prevent the transmission of infectious diseases. However, the frequent use of alcohol-based hand sanitizers may cause skin dryness and irritation, making natural antiseptic alternatives necessary. Mangosteen leaves (Garcinia mangostana L.) contain secondary metabolites with potential antibacterial activity. This study aimed to formulate an ethanol extract of mangosteen leaves into a hand sanitizer gel, evaluate its physical quality, determine its antibacterial activity against Escherichia coli, and identify the most effective extract concentration. This laboratory experimental study used ethanol extract of mangosteen leaves at concentrations of 5%, 10%, and 15%. The formulations were evaluated for organoleptic properties, pH, clarity, viscosity, and drying time. Antibacterial activity was determined using the well diffusion method against Escherichia coli. The data were analyzed using One-Way Analysis of Variance (ANOVA), followed by Tukey's Honestly Significant Difference (HSD) post hoc test. This study showed that all formulations met the physical quality requirements and exhibited antibacterial activity against Escherichia coli. Statistical analysis revealed significant differences among the formulations (p < 0.05). The formulation containing 15% ethanol extract demonstrated the highest antibacterial activity, with a mean inhibition zone diameter of 12.76 mm, which was classified as strong. In conclusion, ethanol extract of mangosteen leaves can be formulated into a hand sanitizer gel with acceptable physical characteristics and antibacterial activity against Escherichia coli, with the 15% extract concentration being the most effective formulation. The suggestion of this study is to develop other dosage forms, evaluate antibacterial activity against different bacterial species, and investigate the long-term stability and viscosity of the formulation.
