Abstract Chantika, Ade Aulia. Characteristics of Secondary Metabolites in Ethanolic Extract of Mangrove Leaves (Rhizophora mucronata L.) and Their Antibacterial Activity Against Streptococcus mutans and Porphyromonas gingivalis Using Thin Layer Chromatography (TLC) Profile Method. Undergraduate Thesis, Department of Pharmacy, Faculty of Pharmacy, Deli Husada Institute of Health, Deli Tua. (Supervised by: Firdaus Fahdi, M.Pd.) Rhizophora mucronata L. leaves are one of the mangrove plants containing various secondary metabolites, including flavonoids, alkaloids, tannins, saponins, and steroids, which have the potential to serve as natural antibacterial agents in response to the increasing problem of antibiotic resistance. This study aimed to evaluate the characterization of the crude drug (simplisia), identify the secondary metabolites present in the ethanol extract of Rhizophora mucronata L. leaves based on Thin-Layer Chromatography (TLC) profiles, and determine its antibacterial activity against Streptococcus mutans and Porphyromonas gingivalis. This study employed an in vitro experimental design. The ethanol extract of Rhizophora mucronata L. leaves was obtained by maceration using 70% ethanol, followed by crude drug characterization, identification of secondary metabolites using TLC, and evaluation of antibacterial activity using the disc diffusion method at concentrations of 50%, 75%, and 100%. The crude drug characterization met the required quality standards, while TLC analysis confirmed the presence of flavonoids, alkaloids, tannins, saponins, and steroids. The ethanol extract of Rhizophora mucronata L. leaves inhibited the growth of Streptococcus mutans and Porphyromonas gingivalis, with the largest inhibition zones observed at the 100% concentration, measuring 16.50 mm and 17.65 mm, respectively. The results of the One-Way ANOVA test indicated a statistically significant difference among the treatment groups (p < 0.05). It can be concluded that the ethanol extract of Rhizophora mucronata L. leaves fulfilled the crude drug characterization requirements, contained various secondary metabolites as identified by TLC profiling, and exhibited antibacterial activity against Streptococcus mutans and Porphyromonas gingivalis, with the 100% concentration showing the highest antibacterial effectiveness.
