Abstract Mangkok leaves (Polyscias scutellaria (Burm. f.) Fosberg) contain secondary metabolites, including alkaloids, flavonoids, saponins, tannins, and steroids, which have potential antibacterial activity. The incorporation of mangkok leaf extract into paper soap is expected to provide a practical alternative soap that is effective in inhibiting the growth of Staphylococcus aureus. This study is aimed to formulate paper soap containing ethanol extract of mangkok leaves (Polyscias scutellaria (Burm. f.) Fosberg) and to determine its antibacterial activity against Staphylococcus aureus in vitro. This study was an experimental study. Ethanol extract of mangkok leaves was formulated into paper soap at concentrations of 40%, 50%, and 60%. The quality evaluation included organoleptic properties, pH, foam height, weight uniformity, and washing time. Antibacterial activity was tested using the disc diffusion method against Staphylococcus aureus. The results showed that all formulations met the requirements for physical quality evaluation. The pH values ranged from 7.25–7.63, foam height ranged from 4.3–4.6 cm, all formulations met the requirements for weight uniformity, and washing time ranged from 10.99–12.76 seconds. The antibacterial activity test showed that the 40%, 50%, and 60% extract formulations produced inhibition zone diameters of 18.40 mm, 19.31 mm, and 20.43 mm, respectively, while the positive control produced an inhibition zone of 22.15 mm. Based on the classification of antibacterial inhibition activity, all formulations were categorized as having strong antibacterial activity, with the highest antibacterial activity observed in the formulation containing 60% extract. Based on the results, it can be concluded that ethanol extract of mangkok leaves (Polyscias scutellaria (Burm. f.) Fosberg) can be formulated into paper soap that meets the requirements for physical quality evaluation and exhibits strong antibacterial activity against Staphylococcus aureus. 1
