| Abstract | Free radicals are molecules that have one or more unpaired electrons that can trigger cell damage. One of the natural sources of antioxidants that has the potential to ward off free radicals is neem leaves (Azadirachta indica A. Juss) because they contain secondary metabolite compounds such as flavonoids, alkaloids, tannins, saponins, and steroids that have the potential to be natural antioxidants. To determine the antioxidant activity and determine the IC valueof 50 ethanol extract of neem leaf (Azadirachta indica A. Juss) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. This study was an experimental study. Neem leaf simplicia is extracted by the maceration method using 96% ethanol. Antioxidant activity testing was carried out using the DPPH method using Uv-Vis spectrophotometry. Method validation included tests of linearity, precision, accuracy, detection limits (LOD), and quantity limits (LOQ). Neem leaf ethanol extract showed antioxidant activity with an IC50 value of 59.54 ppm, so this extract included in the category of strong antioxidants. The results of the method validation show that the parameters of linearity, precision, accuracy, LOD, and LOQ have met the method validation requirements. Neem leaf ethanol extract (Azadirachta indica A. Juss) has strong antioxidant activity against DPPH free radicals with an ICvalue of 50 of 59.54 ppm. The analysis method has fullfilled the validation requirements so that it is suitable used in determining antioxidant activity. Future research is recommended to use other methods, such as ABTS or FRAP, to evaluate the antioxidant activity of neem leaves (Azadirachta indica A. Juss.). | 1 |
