| Abstract | Neem leaves (Azadirachta indica A. Juss.) are medicinal plants that contain various secondary metabolites, including flavonoids, alkaloids, tannins, and saponins. Flavonoids are known for their antioxidant activity and have the potential to prevent diseases caused by free radicals. Therefore, determining the total flavonoid content is important as one of the quality parameters of herbal extracts. This study aimed to qualitatively identify flavonoids, determine the total flavonoid content of the ethanolic extract of neem leaves, and validate the analytical method using UV-Visible spectrophotometry. This study was an experimental laboratory study. Neem leaf simplicia were extracted by maceration using 96% ethanol as the solvent. Qualitative analysis was performed through phytochemical screening. The total flavonoid content was determined using the aluminum chloride (AlCl₃) colorimetric method with quercetin as the reference standard and measured using a UV-Visible spectrophotometer. Method validation included linearity, accuracy, precision, limit of detection (LOD), and limit of quantification (LOQ).The results showed that the ethanolic extract of neem leaves tested positive for flavonoids based on phytochemical screening. The quercetin calibration curve produced the regression equation y = 0.06641x + 0.03775 with a coefficient of determination (R² = 0.9903), indicating a good linear relationship between concentration and absorbance. The average total flavonoid content of the ethanolic extract of neem leaves was 4.5412%. Method validation showed an accuracy (% recovery) of 96.87%, a precision (%RSD) of 1.8850%, an LOD of 1,216 mg/L, and an LOQ of 3,685 mg/L. These results indicate that the UV-Visible spectrophotometric method met the validation requirements and is suitable for determining the total flavonoid content of the ethanolic extract of neem leaves. Further research is recommended to use other analytical methods, such as High-Performance Liquid Chromatography (HPLC), so that the flavonoid content obtained can be compared with the UV-Vis spectrophotometric method to obtain more comprehensive data | 1 |
