| Abstract | Acne is one of the most common skin disorders caused by various factors, including bacterial infection by Propionibacterium acnes. The utilization of natural products as antibacterial agents has gained increasing attention, one of which is Singkil leaf (Premna cordifolia Roxb.), known to contain secondary metabolites such as alkaloids, flavonoids, tannins, saponins, and steroids with potential antibacterial properties. This study aimed to formulate ethanol extract of Singkil leaves into a cleansing balm preparation, evaluate its physical quality, and determine its antibacterial activity against Propionibacterium acnes. This study was conducted using an experimental laboratory method. The ethanol extract of Singkil leaves was formulated into cleansing balm preparations at concentrations of 10%, 15%, and 20%. Physical evaluations included organoleptic, homogeneity, pH, spreadability, and viscosity tests. Antibacterial activity was assessed using the agar well diffusion method against Propionibacterium acnes with three replications. Data were analyzed using the One-Way ANOVA test. The results showed that all formulations had the required physical quality standards, exhibiting good homogeneity, pH values ranging from 4.5 to 7.0, spreadability of 5–7 cm, and appropriate viscosity. The average inhibition zone diameters at extract concentrations of 10%, 15%, and 20% were 14.56 mm, 15.28 mm, and 17.21 mm, respectively. The positive control produced an inhibition zone of 18.18 mm, while the negative control showed no inhibition zone. All formulations demonstrated strong antibacterial activity, with the highest activity observed at the 20% concentration.study was the cleansing balm containing ethanol extract of Singkil leaves (Premna cordifolia Roxb.) fulfilled the required physical quality standards and exhibited antibacterial activity against Propionibacterium acnes. The 20% extract concentration was identified as the most effective formulation in inhibiting bacterial growth. Further studies are suggested to evaluate the long-term stability of the formulation, investigate its activity against other acne-causing bacteria, and conduct safety and efficacy tests on human volunteers to support its development as a natural anti-acne cosmetic product.
