| Abstract | Wati, Nur Nila. Phytochemical Analysis and Determination of Total Alkaloid Content of Ethanolic Extract of Petai Cina Leaves (Leucaena leucocephala (Lam.) de Wit) Using UV-Visible Spectrophotometric Method. Thesis, Department of Pharmacy, Faculty of Pharmacy, Deli Husada Institute of Health, Deli Tua, in 2026. (Supervised by: apt. Sulasmi, S.Farm., M.KM., M.Farm). Petai cina leaves (Leucaena leucocephala (Lam.) de Wit) are known to contain various secondary metabolites, including alkaloids, which have the potential to be developed as medicinal compounds. This study aimed to analyze the phytochemical constituents and determine the total alkaloid content of the 70% ethanolic extract of petai cina leaves using the UV-Visible spectrophotometric method, as well as to evaluate the validity of the analytical method. The study was conducted experimentally. The leaves were extracted by the percolation method using 70% ethanol. Phytochemical screening was performed, and the total alkaloid content was determined using bromocresol green (BCG) reagent with caffeine as the reference standard at a wavelength of 416 nm. The analytical method was validated through linearity, accuracy, precision, limit of detection (LOD), and limit of quantification (LOQ) tests. This study showed that the 70% ethanolic extract of petai cina leaves contained alkaloids, flavonoids, saponins, tannins, and steroids, with a total alkaloid content of 65.43%. The validation results demonstrated that all validation parameters met the required acceptance criteria, indicating that the UV-Visible spectrophotometric method was valid for determining the total alkaloid content. It can be concluded that the 70% ethanolic extract of petai cina leaves contains alkaloids that can be determined accurately using the validated UV-Visible spectrophotometric method. The suggestion of this study is was future researcher should investigate different extraction solvents or analytical methods, such as HPLC or FTIR, and further explore the potential of petai cina leaves as a raw material for pharmaceutical preparations or herbal medicinal products.
