| Abstract | Dental caries is one of the most common oral diseases, primarily caused by Streptococcus mutans. Purple passion fruit seeds (Passiflora Edulis Sims) contain secondary metabolites such as flavonoids, tannins, alkaloids, and saponins, which have potential antibacterial activity. This study is aimed to evaluate the effectiveness of a mouth spray formulation containing ethanol extract of purple passion fruit seeds against Streptococcus mutans and to determined the most effective extract concentration. This laboratory experimental study employed a post-test only control group design. The ethanol extract of purple passion fruit seeds was formulated into mouth spray preparations at concentrations of 5% (F1), 10% (F2), and 15% (F3). The formulations were evaluated for organoleptic properties, homogeneity, pH, viscosity, specific gravity, and stability. Antibacterial activity was assessed using the diffusion method against Streptococcus mutans. Data were analyzed using normality and homogeneity tests, followed by One-Way ANOVA and Tukey HSD post hoc test at a 95% confidence level. The results demonstrated that all mouth spray formulations met the required evaluation standards. The mean inhibition zone diameters were 10.69 mm, 10.93 mm, and 12.68 mm for F1, F2, and F3, respectively, while the positive control produced an inhibition zone of 16.38 mm. One-Way ANOVA indicated a statistically significant difference among the treatment groups (p<0.05). The formulation containing 15% ethanol extract (F3) exhibited the highest antibacterial activity against Streptococcus mutans. | 1 |
