Abstract Hulu, Gilbert Stevan Memories. “The Formulation of a Gel Preparation Containing Red Andong Leaf Ethanol Extract (Cordyline fruticosa (L.) A.Chev.) for Incision Wound Healing in Male White Rats (Rattus norvegicus).” Thesis, Department of Pharmacy, Faculty of Pharmacy, Deli Husada Deli Tua Institute of Health. (Supervised by: apt. Ftiri Wahyu Dhini, S.Farm., M.K.M.) Incision wounds require appropriate management to accelerate healing and prevent complications. Red dracaena leaves (Cordyline fruticosa (L.) A.Chev.) contain flavonoids, alkaloids, tannins, saponins and steroids that may support wound healing and were therefore formulated into a gel. This study is aimed to formulate an ethanolic extract of C. fruticosa into a gel, evaluate its physical quality and assess its effectiveness in treating incision wounds in male white rats. An experimental design was employed, including plant identification, simplisia preparation, ethanolic (96%) maceration extraction and phytochemical screening. Gels were prepared with extract concentrations of 5%, 15%, and 25%, along with gel base (negative control) and Bioplacenton gel (positive control). Physical quality evaluation included organoleptic properties, homogeneity, pH, spreadability and viscosity. Wound healing effectiveness was assessed by measuring wound length daily for 7 days and analyzed using One-Way ANOVA followed by Tukey HSD test. Results confirmed plant identity as C. fruticosa with 15% extract yield and positive phytochemical screening for flavonoids, alkaloids, tannins, saponins, and steroids. All gel formulas met physical quality requirements (pH 6–7; spreadability 5.61–5.94 cm; viscosity 13,733–44,000 cP). The highest wound healing percentage was observed in Bioplacenton gel (51.2%), followed by 25% extract gel (41.3%), 15% (37.0%), 5% (33.0%), and gel base (32.5%). Statistical analysis showed a highly significant difference among groups (p = 0.000), with higher extract concentrations associated with faster healing. Ethanolic extract of red dracaena leaves can be formulated into a gel meeting physical quality standards and effectively accelerates incision wound healing, with 25% concentration showing the best effect. 1
