Abstract Hand hygiene is one of the important efforts to prevent the spread of infectious diseases caused by bacteria, such as Staphylococcus aureus and Escherichia coli. One natural ingredient that has potential as an antibacterial agent is bandotan leaf (Ageratum conyzoides L.) because it contains secondary metabolites with antibacterial activity. This study aimed to formulate ethanol extract of bandotan leaves into a handwash preparation, evaluate the physical quality of the preparation, and determine its antibacterial activity against Staphylococcus aureus and Escherichia coli. This study was an experimental study using ethanol extract of bandotan leaves obtained through maceration using 96% ethanol. The extract was formulated into handwash preparations at concentrations of 10%, 20%, and 30%. The resulting preparations were evaluated based on organoleptic properties, homogeneity, pH, foam height, and viscosity. Antibacterial activity was tested using the disk diffusion method against Staphylococcus aureus and Escherichia coli. The results showed that all handwash formulations containing ethanol extract of bandotan leaves met the physical evaluation requirements. The handwash preparations also showed antibacterial activity against Staphylococcus aureus and Escherichia coli. Against Staphylococcus aureus, the average inhibition zone diameters of F1 (10%), F2 (20%), and F3 (30%) were 18,76 mm, 19,26 mm, and 20,70 mm, respectively, with a strong category. Against Escherichia coli, the average inhibition zone diameters of F1 (10%), F2 (20%), and F3 (30%) were 15,76 mm, 16,70 mm, and 18,80 mm, respectively, with a strong category. The results showed that increasing the concentration of ethanol extract of bandotan leaves increased the antibacterial activity against both test bacteria. Formula F3 with an extract concentration of 30% showed the best antibacterial activity. Based on the results of the study, it can be concluded that ethanol extract of bandotan leaves (Ageratum conyzoides L.) was successfully formulated into a handwash preparation that met the physical evaluation requirements and showed antibacterial activity against Staphylococcus aureus and Escherichia coli. Increasing the extract concentration increased the antibacterial activity against both test bacteria, with F3 at a concentration of 30% providing the best antibacterial activity. 1
