| Abstract | Sari, Intan. "Antioxidant Activity of the Extract and Fraction of Knobweed Leaves (Hyptis capitata Jacq.) and Formulation of Knobweed Leaf Ointment against Staphylococcus aureus." 2026. Undergraduate Thesis, Department of Pharmacy, Faculty of Pharmacy, Institut Kesehatan Deli Husada Deli Tua. Supervised by apt. Novarianti Marbun, S.Farm., M.Si. Skin infections caused by Staphylococcus aureus remain a growing public health problem due to the increasing prevalence of antibiotic resistance. Knop grass (Hyptis capitata Jacq.) leaves are known to contain secondary metabolites, including flavonoids, alkaloids, tannins, saponins, and steroids, which have potential antioxidant and antibacterial activities. This study aimed to determine the antioxidant activity of the ethanol extract and ethyl acetate fraction of Hyptis capitata Jacq. leaves, as well as to evaluate the antibacterial activity of ointment formulations against Staphylococcus aureus. This experimental study involved the preparation of simplicia, extraction, fractionation, antioxidant activity testing using the 2,2 diphenyl-1-picrylhydrazyl (DPPH) method, formulation of W/O and O/W ointment bases, evaluation of the physical quality of the ointments, and antibacterial activity testing using the agar diffusion method. The results showed that the ethyl acetate fraction exhibited stronger antioxidant activity than the ethanol extract, as indicated by its lower IC₅₀ value of 86.23 ppm. The formulated ointments had the required physical quality standards and were able to inhibit the growth of Staphylococcus aureus. This finding concluded that the O/W ointment base produced a larger inhibition zone than the W/O base, with an average inhibition zone diameter of 25.8 mm (classified as very strong), indicating that the release of the active compounds from the O/W base was more optimal. It is suggested that further research conduct in vivo studies and skin irritation testing to evaluate the safety and clinical efficacy of this ointment formulation.
