| Abstract | Acne (Acne vulgaris) is a skin inflammation of the pilosebaceous unit commonly triggered by Cutibacterium acnes, where long-term use of conventional antibiotics poses a risk of resistance. This experimental study aimed to formulate, evaluate the physical properties, and test the antibacterial activity of a gel combination containing papaya seed ethanol extract (Carica papaya L.) and Virgin Coconut Oil (VCO) against Cutibacterium acnes. The extract was obtained via maceration using 96% ethanol, yielding 10.173% and confirming the presence of alkaloids, flavonoids, saponins, tannins, and steroids. Gel formulations with varying active ingredient concentrations (F1 0%:2.5%, F2 2.5%:5%, F3 5%:7.5%, F4 7.5%:10%, and F5 10%:0%) were evaluated over 3 weeks of storage for organoleptic properties, homogeneity, pH (5.08–5.83), spreadability (5.35–6.42 cm), and viscosity (3,347–30,720 cPs), all of which met pharmaceutical quality standards. Antibacterial testing using the disc diffusion method showed inhibition zones across all formulations, with Formula F4 (7.5% papaya seed extract and 10% VCO) exhibiting the highest activity with an average inhibition zone diameter of 16.1 mm. Thus, the combination of papaya seed ethanol extract and VCO can be formulated into a physically stable gel preparation with strong antibacterial activity against acne-causing bacteria. | 1 |
