| Abstract | Acne is a common skin problem experienced by many people approximately 40%–80% in Southeast Asia and 80%–85% in Indonesia among those aged 15–18 years. The bacterium responsible for acne is Propionibacterium acnes, which plays a role in skin inflammation. One plant with potential as an anti-acne agent is Chinese betel leaf (Peperomia pellucida), which is known to contain secondary metabolites such as alkaloids, flavonoids, saponins, and tannins that exhibit antibacterial potential. This study aimed to formulate a Chinese betel leaf nanosimplicia into a peel-off mask preparation, to determine the effect of varying sonication time on the characteristics of the nanosimplicia and its antibacterial activity against Propionibacterium acnes, and to compare its effectiveness with peel-off masks on the market. The study was conducted experimentally with varying sonication times of 10, 20, and 30 minutes at a concentration of 5%. Data were then analyzed using a One-Way ANOVA test to determine significant differences in nanosimplicia characterization, preparation evaluation, and antibacterial activity. The results show that Characterization of the nanosimplisia formulations showed that the smallest particle size was obtained after 30 minutes of sonication, at 149.20 nm, with a polydispersity index of 0.19 and a zeta potential of 34.17 mV. All formulations met the physical evaluation requirements for peel-off masks. Antibacterial activity testing showed that the mask base produced no inhibition zone (0 mm), whereas the nanosimplisia formulations sonicated for 10, 20, and 30 minutes produced average inhibition zone diameters of 11.55 mm, 15.03 mm, and 18.37 mm, respectively, while the positive control (Favbeauty acne) had an inhibition zone diameter of 16.67 mm. Statistical analysis showed that variations in sonication time had a significant effect on the characteristics of the nanosimplisia and antibacterial activity (p<0.05). The conclusion show that A nanosimplisia peel-off mask made from Chinese betel leaf has been successfully formulated and exhibits antibacterial activity against Propionibacterium acnes. The suggestion from this study show that further research should be conducted to evaluate the stability, optimize the nanosimplisia concentration, and assess the irritation potential and in vivo efficacy of the formulation. | 1 |
